白介素-1β对脓毒症幼鼠海马SNAP-25表达的影响

摘要: 目的 探讨白介素-1β(IL-1β)对脂多糖(LPS)诱导脓毒症新生幼鼠突触体相关蛋白25(SNAP-25)表达的影响。方法 SD幼鼠随机(随机数字法)分为对照组和脓毒症模型组。用1mg/kg LPS腹腔注射SD P1 d幼鼠诱导脓毒症模型,对照组用等剂量0.01 mol/L磷酸盐缓冲液(PBS)腹腔注射。Western blot检测腹腔注射LPS 1、2和3 d后海马组织IL-1β和白细胞介素1受体1(IL-1R1)的蛋白表达,7、14、28 d后海马组织SNAP-25的蛋白表达。原代海马神经元细胞培养24 h后,分为四组即对照组、IL-1β组(40 ng/mL)、IL-1β(40 ng/mL)+白介素1受体拮抗剂(IL-1Ra)(40 ng/mL)组、IL-1Ra(40 ng/mL)组,干预3 d。CCK-8检测海马神经元活性,NeuN免疫荧光染色鉴定海马神经元纯度,Western blot和Real-time PCR分别检测各组SNAP-25的蛋白和mRNA水平。采用SPSS 22.0统计软件分析,数据分别采用student-t检验、Dunnett-t检验以及析因设计的方差分析,以P < 0.05为差异具有统计学意义。结果 与对照组相比,腹腔注射LPS 1、2、3 d后幼鼠海马组织IL-1β和IL-1R1的蛋白表达明显增加(P < 0.05);7、14、28 d后SNAP-25蛋白表达显著下降(P < 0.05)。免疫荧光NeuN染色阳性细胞数约92%。用IL-1β干预海马神经元24 h后,与对照组相比SNAP-25蛋白水平降低(P < 0.05),用IL-1Ra阻断后可以逆转IL-1β诱导的SNAP-25蛋白低表达(P < 0.05), 而各处理组SNAP-25的mRNA水平差异无统计学意义(P > 0.05)。结论 脓毒症幼鼠海马内释放大量IL-1β可能通过其受体IL-1R1抑制神经元内SNAP-25蛋白表达,这可能是引起脓毒症幼鼠后期学习记忆障碍的重要原因。

Effect of IL-1β on expression of SNAP-25 in the hippocampus in septic neonatal rats

Abstract: Objective To investigate the effect of interleukin-1β (IL-1β) on the expression of synaptic protein SNAP-25 in the hippocampus in septic neonatal rat induced by systemic lipopolysaccharide (LPS) injection. Methods Sprague-Dawley (SD) rats were randomly divided into two groups: control group and sepsis group. The rat model of sepsis was produced by intraperitoneal injection of 1 mg/kg LPS, and rats in the control group were injected with an equal volume of 0.01 mol/L phosphate buffered saline (PBS). The expression levels of IL-1β and IL-1R1 in the hippocampus at 1, 2 and 3 d, and synaptosomal-associated protein 25 (SNAP-25) at 7, 14 and 24 d after LPS intraperitoneal injection were detected by Western blot. After cultured for 24 h, primary hippocampal neurons were divided into four groups including the control group, IL-1β (40 ng/mL) treatment group, IL-1β (40 ng/mL) + IL-1Ra (40 ng/mL) treatment group, and IL-1Ra (40 ng/mL) treatment group. The effect of IL-1β on SNAP-25 expression in primary hippocampal neuron was determined by Western blot and real-time PCR. The purity of hippocampal neurons were identified by NeuN immunofluorescence staining and the activity of neurons were detected by CCK-8 assay. All data were analyzed by SPSS version 22.0. The data were analyzed by student-t test and Dunnett-t test. The interaction effects were analyzed by factorial ANOVA. Differences were considered to be statistically significant if P < 0.05. Results Compared with the control group, the expressions of IL-1β and IL-1R1 were significantly increased in the hippocampus at 1, 2 and 3 d after intraperitoneal injection of LPS (P < 0.05). The expression of SNAP-25 protein was decreased at 7, 14, and 28 d after intraperitoneal injection of LPS (P < 0.05). The purity of primary neurons was about up to 92%. The activity of primary neurons was not relatively changed after treated with IL-1β at a dose less than 40 ng/mL. The level of SNAP-25 protein was obviously decreased in primary neurons at 24 h after IL-1β treatment (P < 0.05). IL-1Ra treatment might reverse the effect of IL-1β on primary neurons (P < 0.05). While, the expression of SNAP-25 mRNA was not statistically different in each group (P > 0.05). Conclusions IL-1β may possibly inhibit the expression level of SNAP-25 protein in the hippocampus in the septic rats through its receptor IL-1R1, which would contribute to cognitive dysfunction of septic neonatal rats in later life.

新生儿脓毒症是威胁新生儿生命的重要原因[1], 在后期会出现认知功能障碍[2],严重影响患儿的生活质量。IL-1β是引起新生儿脓毒症后期认知障碍的重要炎症因子[3]。SNAP-25是可溶性N-甲基马来酰亚胺敏感因子附着蛋白受体复合体的组成成分之一,该复合体参与了突触囊泡的胞吐[4],而在突触传递中起关键作用。IL-1β可导致突触损伤,其中激活的IL-1R信号通路起着重要作用[5-9]。目前还没有研究揭示IL-1β与海马突触蛋白SNAP-25的联系。因此本研究采用LPS诱导脓毒症模型以及IL-1β刺激原代培养的SD幼鼠海马神经元,探讨IL-1β对神经元内SNAP-25蛋白表达的影响。

1 材料与方法 1.1 动物

研究所需的SD大鼠(1日龄)由广州中医药大学实验动物中心提供,许可证号:SCXK(粤)2013-0034。动物被随机(随机数字法)分为对照组和脓毒症模型组,用LPS 1 mg/kg腹腔注射SD P1 d幼鼠诱导脓毒症模型,对照组用等剂量0.01 mol/L磷酸盐缓冲液(PBS)腹腔注射。动物处理后在动物房饲养1、2、3、7、14、28 d用于实验。

1.2 主要试剂

细胞培养所需试剂购自美国Gibco公司;LPS购自美国Sigma公司;IL-1β和IL-1Ra购自美国Proteintech公司;CCK-8试剂盒购自日本DOJINDO公司;除NeuN抗体购自美国Millipore公司,IL-1R1抗体购自美国Santa Cruz Biotechnology公司外,其余抗体购自美国Cell Signaling Technology公司;PrimeScriptTM RTMaster Mix和SYBR® Premix Ex TaqTM Ⅱ购自日本TaKara公司;引物由中国武汉谷歌生物有限公司设计合成。

1.3 主要方法 1.3.1 原代海马神经元培养

使用新生SD大鼠(1日龄)进行原代海马神经元培养。从新生乳鼠脑中分离海马,剔除血管膜,剪碎成大小不超过1 mm3的小组织块,用0.125%胰蛋白酶在37 ℃消化10 min后用FBS终止消化。将细胞轻柔吹打成悬液后200目滤网过滤。取细胞悬液1 100 r/min离心5 min,将细胞重悬在完全培养基内。6 h后换液,将DMEM/F12置换成Neurobasal培养基,另加入2% B27以及1%谷氨酰胺。细胞接种在提前用多聚赖氨酸处理过的6孔板或者96孔板上。

1.3.2 原代细胞药物处理及实验分组

CCK-8测定IL-1β的干预浓度。方法按照制造商的说明书进行。原代培养的神经元(3×103/孔)用不同浓度(0、10、20、40、80、160 ng/mL)的IL-1β在96孔板中培养24 h,然后将10 μL CCK-8工作溶液加入到每个孔中,在37 ℃下温育6 h。用酶标仪检测450 nm波长下每孔的吸光度(A)值。原代细胞以2×106/孔的密度接种在六孔板上。培养24 h后,随机分为对照组(0.01 mol/L PBS)、IL-1β(40 ng/mL)组、IL-1β(40 ng/mL) +IL-1Ra (40 ng/mL)组、IL-1Ra (40 ng/mL)组。

1.3.3 Western blot

使用蛋白提取试剂盒提取海马组织或者原代神经元细胞的总蛋白。BCA法测蛋白浓度。使用10%的分离胶,在200 mA恒定电流下转膜120 min。5%牛奶室温封闭1 h,然后与对应一抗4 ℃孵育过夜。二抗室温孵育1 h。照相显影通过化学发光(ECL发光液)。使用ImageJ程序定量Western blot条带。

1.3.4 细胞免疫荧光及图像分析

原代培养的海马神经元细胞,在PBS中漂洗三次后用4%的多聚甲醛固定15 min后10%驴血清室温封闭1 h。细胞孵育NeuN (1:200)过夜。次日,488驴抗鼠IgG(H + L)室温孵育1 h。DAPI孵育5 min后拍照观察。

1.3.5 Real-time PCR

使用trizol法提取细胞总RNA。使用试剂盒并分别按照说明书进行逆转录以及扩增。引物序列如下:大鼠SNAP-25正向引物序列CGTAATGAACTGGAGATGCA; 大鼠SNAP-25反向引物序列GATCGAGTTGTTCGCCTTGCT; 大鼠β-actin正向引物序列TGCTATGTTGCCCTAGACTTCG; 大鼠β-actin反向引物序列GTTGGCATAGAGGTCTTTACGG。以β-actin作为内参基因,用2-△△CT法对结果进行计算和分析。

1.4 统计学方法

应用SPSS 22.0统计分析软件对数据进行处理, 使用Graphpad Prism 5.0作图,计量资料以均数±标准差(Mean±SD)表示。海马组织蛋白表达数据采用student-t检验,IL-1β处理海马神经元采用Dunnett-t检验。SNAP-25蛋白和mRNA表达的比较采用析因设计的方差分析,若存在交互作用进行单独效应分析;不存在交互作用,如果数据方差齐,则通过LSD多重比较; 否则Dunnett-T3方法分析。以P < 0.05为差异有统计学意义。

2 结果 2.1 海马组织SNAP25的表达

蛋白印迹显示:与对照组相比腹腔注射LPS组在7、14、28 d海马内SNAP-25蛋白表达明显减少且差异有统计学意义(正态性检验各组P > 0.05; 7 d control组vs 7 d LPS组:t = 2.463,P < 0.05;14 d control组vs 14 d LPS组:t = 3.130,P < 0.05;28 d control组vs 28 d LPS组:t = 7.218,P < 0.05),见图 1

2.2 海马组织IL-1β和IL-1R1的表达

蛋白印迹检测海马组织发现:与对照组相比,腹腔注射LPS 1、2、3 d,海马组织IL-1β(正态性检验各组P > 0.05; 1 d control组vs 1 d LPS组:t = 5.114,P < 0.05;2 d control组vs 2 d LPS组:t = 4.811,P < 0.05;3 d control组vs 3 d LPS组:t = 2.888,P < 0.05)和IL-1R1(正态性检验各组P > 0.05,1 d control组vs 1 d LPS组:t = 3.060,P < 0.05;2 d control组vs 2 d LPS组:t = 2.534,P < 0.05;3 d control组vs 3 d LPS组:t = 2.802,P < 0.05)表达明显上调,见图 2

2.3 NeuN染色鉴定神经元纯度

任意取三个视野,计算NeuN染色阳性细胞数。本研究中原代海马神经元纯度为(92.42±0.04)%,见图 3

2.4 IL-1β处理海马神经元

为确定IL-1β的干预浓度,采用CCK-8试剂盒检测给予原代海马神经元不同浓度(0、10、20、40、80、160 ng/mL)IL-1β处理后神经元的活性。结果显示:在IL-1β浓度不超过40 ng/mL时,神经元能保持相对较高的活性(10个vs 0个,P > 0.05;20个vs 0个,P > 0.05;40个vs 0个, P > 0.05);当IL-1β在80和160 ng/mL的浓度时,细胞活性明显下降(80 vs 0, P < 0.05;160 vs 0, P < 0.05),因此选择40 ng/mL浓度的IL-1β处理神经元(正态性检验各组P > 0.05, 方差齐性检验P = 0.121)。见图 4

2.5 IL-1β对海马神经元内SNAP-25表达的影响

使用IL-1β或IL-1Ra处理原代海马神经元后,通过Western blot检测各组SNAP-25蛋白表达。IL-1β与IL-1Ra存在交互效应(F = 19.817,P < 0.05)。进行单独效应分析发现,与control组相比,IL-1β处理24 h后原代神经元内SNAP-25蛋白表达下降(P < 0.05);IL-1β+IL-1Ra组与IL-1β组相比,SNAP-25蛋白水平增加(P < 0.05);而各组间SNAP-25的mRNA水平差异无统计学意义(control组vs IL-1β组:P > 0.05;IL-1β vs IL-1β+IL-1Ra: P > 0.05)。见图 5

3 讨论

脓毒症对中枢神经系统的损伤中炎症因子的参与起了重要作用, IL-1β作为主要的促炎因子在此过程中备受关注[10]。LPS可使小胶质细胞激活并分泌IL-1β,高浓度的IL-1β作用于神经元可以抑制突触强度和LTP[5-6]。IL-1β直接功能主要是通过其受体IL-1R1介导。IL-1R1在海马神经元和星形胶质细胞中均有表达[11-12]。本研究发现:新生脓毒症幼鼠海马内IL-1β大量释放和IL-1R1被激活,并且海马内SNAP-25蛋白表达下调。为了进一步证明IL-1β可以抑制神经元内SNAP-25蛋白水平,本研究体外神经元培养的结果发现:IL-1β处理可以导致原代神经元内SNAP-25蛋白水平降低,用IL-1R1拮抗剂可以逆转IL-1β导致的SNAP-25蛋白的降低,同时各组SNAP-25的mRNA水平差异无统计学意义。

SNAP-25与突触囊泡胞吐关系紧密。SNAP-25是可溶性N-甲基马来酰亚胺敏感因子附着蛋白受体复合体核心蛋白组成成分之一[13]。它不仅可以调节胞吐[14],其表达的异常会导致认知功能障碍和行为异常[15]。突触前膜释放神经递质是突触信息传递的重要环节。谷氨酸作为脑内重要的兴奋性神经递质,在突触传递、学习与记忆功能中发挥着重要的生理功能[16],它过度激活突触后膜N-甲基-D-天冬氨酸受体会导致神经毒性,造成神经元肿胀坏死[17]。本研究发现脓毒症幼鼠的SNAP-25蛋白的表达水平降低,而在笔者未发表的文章中同时发现了脓毒症幼鼠的学习记忆能力的损伤。因此,笔者推测脓毒症幼鼠海马内炎症反应激活免疫细胞产生大量IL-1β可能通过其受体IL-1R1降低神经元内SNAP-25蛋白水平,这可能影响突触囊泡胞吐,进而可能影响神经递质例如谷氨酸的释放,导致突触信息传递的异常,这可能是引起脓毒症幼鼠后期认知功能障碍的重要机制。
